And even more important: What is the reason for the lower allergy rate at farms?<\/p>\n<p>If we now go back to table 1 there is a 5,0 fold Der p1 excess in farm &#8211; much higher than the 1,7 fold increase of endotoxin. As nearly all allergens have helminth homologues &#8211; \u00a0the question is what did the authors really measure? <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25404363\">Da Costa Santiago 2015<\/a> has a nice table that could give an answer.<\/p>\n<figure id=\"attachment_12897\" aria-describedby=\"caption-attachment-12897\" style=\"width: 1040px\" class=\"wp-caption alignnone\"><a href=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5.jpg\" data-rel=\"key-image-2\" data-rl_title=\"\" data-rl_caption=\"\" title=\"\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-12897 size-full\" src=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5.jpg\" alt=\"\" width=\"1040\" height=\"665\" srcset=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5.jpg 1040w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5-620x396.jpg 620w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5-768x491.jpg 768w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-5-782x500.jpg 782w\" sizes=\"auto, (max-width: 1040px) 100vw, 1040px\" \/><\/a><figcaption id=\"caption-attachment-12897\" class=\"wp-caption-text\">J Immunol. 2015 Jan 1;194(1):93-100 shows Der p1 homologs<\/figcaption><\/figure>\n<p>Der p1 is a cysteine protease of 25kDa that has homologues for example in <em>Loa loa<\/em>. Unfortuneately <em>Loa loa<\/em> is not an explanation at Bavarian farms &#8211; as it is <a href=\"http:\/\/www.thelancet.com\/retrieve\/pii\/S1473309913702639\">more common in tropical Africa<\/a>. Cysteine proteases are nevertheless a big tool for helminths &#8211; \u00a0in\u00a0<em>Schistosoma japonicum<\/em>\u00a0a cathepsin B2 cysteine protease is considered the <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC6107106\/\">main penetration tool<\/a>.<\/p>\n<p>Usually <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26433526\">cysteine proteases are not allergenic<\/a>, but the excessive (and rather isolated) rise combined with a reduced Der \u00a0p1 sensitization in the children, \u00a0is definitely an unusual finding. The Dermatophagoides pteronyssinus habitat is cosmopolitan, house dust,\u00a0 also influenced by altitude, but there is no known reasons for this excess in farms. Is the Der p1 value just a false positive and has it helminthic and not mite origin?<\/p>\n<p>I am therefore blasting Der p1 \u00a0sequence (<a href=\"https:\/\/www.uniprot.org\/uniprot\/P08176\">P08176<\/a>) against all known cow endoparasite genomes. <a href=\"https:\/\/parasite.wormbase.org\/Fasciola_hepatica_prjna179522\/Info\/Index\/\">Fasciola has a strong hit<\/a> with CL6, a <a href=\"https:\/\/www.uniprot.org\/uniprot\/A0A2H1BX03\">papain family cysteine protease<\/a> with e+48.<\/p>\n<figure id=\"attachment_12933\" aria-describedby=\"caption-attachment-12933\" style=\"width: 1152px\" class=\"wp-caption alignnone\"><a href=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9.png\" data-rel=\"key-image-3\" data-rl_title=\"\" data-rl_caption=\"\" title=\"\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-12933 size-full\" src=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9.png\" alt=\"\" width=\"1152\" height=\"722\" srcset=\"https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9.png 1152w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9-620x389.png 620w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9-768x481.png 768w, https:\/\/www.wjst.de\/blog\/wp-content\/uploads\/2019\/07\/screen-9-798x500.png 798w\" sizes=\"auto, (max-width: 1152px) 100vw, 1152px\" \/><\/a><figcaption id=\"caption-attachment-12933\" class=\"wp-caption-text\">Wormbase blastp results &#8211; e+ values are somewhat lower than my local blast result at the same locus<\/figcaption><\/figure>\n<p>With identity values between 40% and 70% there could well be a confusion of mite Der p1 and\u00a0 helminth (Fasciola?) CL6, which should be tested for cross-reactivity.<\/p>\n<p>The story even gets more interesting <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16210902\">when reading more about parasites<\/a><\/p>\n<div class=\"page\" title=\"Page 9\">\n<div class=\"section\">\n<div class=\"layoutArea\">\n<div class=\"column\">\n<blockquote><p>Helminth parasites have complicated life cycles &#8230; at the same time as skewing the immune system toward a Th2-driven response, they have a general suppressive effect on the host immune system that prevents their elimination and reduces immune-mediated tissue damage. It has been suggested that cytokines of the anti-inflammatory network, particularly IL-10 and transforming growth factor- (TGF-), that are produced in response to continual stimulation of the immune system by parasite antigens, are pivotal to regulating the damage they cause and that, coincidentally, these have a bystander protective affect against allergic reactions.<\/p><\/blockquote>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<p>This is exactly what <a href=\"https:\/\/www.nejm.org\/doi\/10.1056\/NEJMoa020057?url_ver=Z39.88-2003&amp;rfr_id=ori:rid:crossref.org&amp;rfr_dat=cr_pub%3dwww.ncbi.nlm.nih.gov\">N Engl J Med 2002; 347:869-877<\/a> showed: increasingly exhausted IL10 capacity.
